Tesamorelin is a synthetic analogue of human growth-hormone-releasing hormone (GHRH 1-44) with a trans-3-hexenoic acid group on the N-terminus that protects it from dipeptidyl-peptidase cleavage. It stimulates pulsatile, physiological GH release from the pituitary.
In a 26-week phase 3 trial in HIV-associated lipodystrophy (n = 412), tesamorelin reduced visceral adipose tissue by 15.2% versus a 5.0% increase with placebo, with corresponding increases in IGF-1 and improvements in triglycerides 1.
